The problem is solved, there were some points along the route with the same M-Value. ArcGIS completely skips those points for calibration, and deviates from the original line. The solution was to remove the point with identical M-values and recalibrate. So it was a curve with real dense points and no Bezier curve.

If you project a lat/long coordinate which is greater than 60 degrees east in the UTM40N zone, it is possible to get a 7 digit Easting. Project to the correct UTM zone to get proper 6 digit coordinates.

Like any projection, Web Mercator has inherent limitations. First, you will notice that no data is cached at the North or South Pole. This is a limitation of all Mercator projections. Also, area and distance measurements will not be correct since the farther north you take the measurement, the more distortion there is. An example is in the picture below, a tissot indicatrix, showing the distortion in projected area.

Problem solved, I forgot to increase the M-resolution for the line feature itself. I only increased the resolution for the calibration points!! Thanks for the help!!

Yes M resolution is 0.0001, tolerance is 0.001 If I increase tolerance/resolution, the problem stays.

You probably projected coordinates outside the valid range of UTM40N. The 'Valid' range of UTM 40 N is approx.from 54 to 60 degrees east. If the projected coordinate is much below 54 degrees, then you can get a negative value. If it is way above 60 degrees you can easily get a 7 digit easting.

Sorry, that's not it. All points are ascending along the line. If you look at the image in my previous reply you will see the calibration points increase, while the hatches around them all show KP 0.000.

I don't think so.. I set the hatches to show every 250 meters. The M-values are KP's (Kilometre Points), and nearly none of them are a multiple of .250 km. Some of the multiple hatches show at a line where I only have one calibration point in their direct proximity. See screenshot below. The red dots presents the values for calibration.
