get zonal statistics from points rather than a raster

I have 3 cruise tracks with many parameters of ocean chemistry data. I want to compare aggregate statistics (mean, mode, SD) of cells across the 3 different cruises;  my idea was to create a fishnet for each sampling session, get the mean value for each parameter within the fishnet polygon, and then compare the polygons. I'm having trouble though with the sampling-- I don't see how to sample from the points to get values for the fishnet polygons. Zonal statistics is almost the right tool but it's meant to get aggregate values from a raster rather than points.

Is there anything wrong with using point to raster and then zonal statistics? Is there a way I could get all the parameters quickly? I feel there's also a way to do this with a spatial join but I'm not sure how.
