Can you explain more what you mean? Do you want your map book to create all pages at one scale? Or will each page be at a different scale depending on the features you are displaying? If you haven't already, look at the tool box tools under Cartography Tools -> Data Driven Pages -> Grid Index Features. You may need to run that tool first in order to create an index layer.
