Is Plan Route available on ArcGIS Desktop? Or just ArcGIS
online?

The Plan Routes tool specifically is only implemented in ArcGIS Online.  If you wanted to accomplish something similar in ArcGIS Desktop you could use Network Analyst ArcGIS Network Analyst | Overview
