Hi everyone,
I have a poster sized map.  The following screenshot shows what a portion of it looks like at 200% zoom in layout view.
The green and red points represent sites that have been sampled, and the grey represent sites which have not yet been sampled.  The labels of the sampled sites show which samples exactly have been taken, there are 4 different kinds.  The points are stored in a file geodatabase, I created labels for them using an advanced expression and converted them to feature-linked annotation.  They are supposed to have line callouts like this: but these aren't showing up.

I was hoping that by converting the labels to annotation I could:
a) Drag the labels to an appropriate location
b) By dragging them, have the missing, or overlapping labels show up
c) By dragging them, have the line callout appear.

b and c are not happening.

My second problem is moving the actual labels.  I want to be able to edit them in layout view but it doesn't look like I can do that.  If I go to data view, then I have to zoom way in to be able to get to the appropriate label viewing scale and by that point, my points are tiny, so I don't get to see what the actual map would look like.  So how am I supposed to place the labels correctly?

Any advice is appreciated, thank-you.

I managed to solve most of the issues.  The data frame was in focus, my problem was that I didn't have the editor "select" tool selected.  I had the "Select" tool from the drawing toolbar and the main toolbar, but it was the editor one that I needed.
My issue with labels not showing up was to do with the advanced expression code I had used.  Somewhere along the road, the symbols I had gotten from the character map got changed and I had to replace them.  Now all the labels are showing.
The issue I am having now is with the line callouts.  When I move the label, the line callout moves with it as if the whole thing was a graphic (see selection in attached image), it doesn't remain pointing to the feature.  I think I may have to re-create the annotations without the line callouts, because it looks as though this functionality is just not available with annotations.

I don't know about your callout questions, but you should be able to edit the anno from layout view. To do that you have to focus the data frame they reside in by either double-clicking the data frame on the layout or by clicking the focus data frame button on the layout toolbar. You should see a thick diagonally hatched outline around the data frame when it is focused.
