Has any one else had issues using web appbuilder (developer addition) in a corporate environment. Have read the help and forum posts and cannot get past the
"Specify the URL to your organization or Portal for ArcGIS" page, getting a
"Unable to access https://xxxxxxx.maps.arcgis.com
A server with the specified hostname could not be found"
Error, any ideas?

Check with your IT staff and see if your firewall is blocking that URL.
I have the same problem when I run with windows service. I, however, able to access AGOL with batch file

Check your browsers network communication to see if anything is returning with a four hundred something error. Also you can check the [install dir]/server/logs/appbuilder.log
What does the error imply (assuming it does returns)

Well different things depending on the number (i.e 403 error means that you to not have the appropriate privileges, 404 means "not found", etc, etc)
I do see 403. How ever, here the problem is I can run batch file. I have temporarily addressed my problem by creating a windows service (on .net framework) and running the batch file in hidden mode. It is working this way. I will give more feedback once I get more information on the problems we are facing.

If your AGOL org name is "myorg", you can directly input URL "https://myorg.maps.arcgis.com" with https protocol into browser's address bar to see whether you can access your org or not. If you can't access your org by browser, it means you can't acess AGOL maybe because of proxy settings or firewall.
this error is due to SSL , if you are using Self Signed Certificate you wont be able to connect to the web app builder , but if it is the domain certificate you will be able to do so. this is works with me find when i used the Domain Certificate
