I recently upgraded my iPad to iOS 10 and noticed the "My Location" button is not working on my WAB app.

I am seeing the same.
From a conversation I had with Esri a few days ago about this issue....
Safari and Chrome now require websites be accessed over https to be able to request a user's location. This has been the case for a little while with Chrome, but is new to Safari. We have this documented for Chrome, but not Safari: https://developers.arcgis.com/javascript/3/jsapi/locatebutton-amd.html That said, a notification should show to alert the user of this restriction.
Please confirm that the locator widget works on your mobile browsers over https. You can use the following web app, in case you do not have SSL enabled on your server: http://arcg.is/2crOHOA.
https works for chrome but the location widget still doesn't work on iOS 10 for me.
On your device with iOS 10, are you using Chrome or Safari? Make sure you enable location services under Settings > Privacy > Location Services > Chrome/Safari (Set to While Using).
I can confirm on iOS 10.0.1 the location widget works on both Chrome and Safari.

Is your WAB app using https protocol?
Yes, https only.
The issue is iOS 10 and standard http protocol.
I contacted ESRI and this issue will be logged as a bug soon.
