do you see any warnings in the web browser about not secure, SSL, etc.?

if you can login into ArcGIS Server Manager or Admin endpoint and portal's Admin endpoint, take a look at the web adaptor settings, federation settings, etc. to see if they look like you expect

does the ArcGIS Server WebContextURL and Portal's WebContextURL value look good too?

If so, they this curveball, on the web server VM, in an administrator command prompt type GPUPDATE /FORCE
