I would like to have the same abilities to style text in ArcGIS Desktop labels and text boxes, including margins and padding, column/row alignment, background colors and images, shadow effects, indentation, etc.  

I want to define a stylesheet somewhere in the document properties, or the layer's label properties, add the appropriate markup to the text label, and then have access to any of the stylizing available in CSS3.

This enhancement would solve a genuine need, and it would make 1:M labeling look really good.

There are no plans to use HTML and CSS in labels. The added complexity, performance, and security implications of essentially having a web page per label make this difficult. If you're just looking for table support, I'd recommend following this issue: https://community.esri.com/t5/arcgis-pro-ideas/create-tabular-labels-with-multiple-attribute/idi-p/9...
