Currently, in ArcGIS Pro Layouts, it is not possible to dynamically link the map scale of one map frame to another. We propose an enhancement that allows users to define the scale of one map frame relative to another Use Case: We create print templates for web maps used at many different map scales. Each layout contains a Detail Map and an Overview Map, where the overview should always show an area about 20× smaller in scale than the detail map (e.g., 1:10,000 → 1:200,000). Suggestion: Add an option in map frame properties to link scales between frames using a ratio or formula

Another use case for project-wide variables and being able to use them in an equation. 
Set 'scaleMapFrameA = 25000' and 'scaleMapFrameB = scaleMapFrameA*10' as example. 

If you have time for a hobby grab Fusion 360 and play around with the variables etc in it.
