I’ve had search online looking to see if ArcGIS Server supports SMB multichannel for a Azure file share or if it’s a single threaded application but wasn’t able to see anything. If we enabled SMB multichannel on a Azure file share does anyone know if ArcGIS Server be able to take advantage of that for better throughput?

ArcGIS Server uses multiple threads, processes, and files during its running operations, so should see a performance gain when using SMB multichannel on a client that is compatible and an Azure Files Premium share. The files that ArcGIS Server uses are typically smaller than 16Kb, so may not see as significant a gain as mentioned in the below documentation.

Hi - Just as a FYI to this topic - we implemented multichannel on our SMB with less than positive results - ArcGIS Server became quite unstable and we ended up rolling the change back.
Would you mind elaborating on the behavior you observed? When I tested on an ArcGIS Server site (albeit with only a few services published and using copied data to the root directories) I saw slightly increased throughput in my results and didn't experience any stability issues.
