Where does the data reside when a hosted feature layer is created on ArcGIS Enterprise?

When a hosted feature layer is created on ArcGIS online, the data goes with it, stored within ArcGIS Online. When you create a hosted feature layer on ArcGIS Enterprise, does the data get stored on the Datastore? Is this the only place the data can be reside?

Correct. The data "behind" hosted feature layers created in the ArcGIS Enterprise portal are stored in the ArcGIS Data Store. And yes, this is the only place they can reside - as the data needs to be managed by the portal. To learn more, please see this whitepaper
