Yes, assuming you mean something like connecting via ArcCatalog. However, you will need to make an administrative connection. See:  http://resources.arcgis.com/en/help/main/10.1/index.html#//0154000003qn000000

Actually I may be wrong about that. I know there are issues if using ArcSDE where you need to upgrade the database. But you should be able to connect from 10.0 to 10.1 server

Yes, I was wrong, can't make an Admin connection in 10.0 to 10.1 but can make a "Use GIS Services" connection
