Network Analyst Route solver lets you set the time of day that you are leaving your start stop and uses predictive or real-time traffic feed to find the shortest path to your destination. Once the solve is done it tells you the arrive time at the destination. That is your ETA.
As far as your second question, "how you can use ArcGIS Network analyst in real time transportation with closest routing to destination:, what is closest routing to destination? There is a closest facility solver that will find the closest facility to a destination and it can use the predictive or real-time traffic to find the shortest path.
