When I publish Indoors feature layers (occupants, events, details, levels, etc.) to my Enterprise portal (referencing registered data), do I need to define the floor field in the layer properties before I publish?

Once you add them to the web map viewer, you can configure the floor-aware of those published feature layers.
In addition, you'll be required to have FACILITIES and LEVELS layers in the map viewer to set up the floor-aware.
