Tried to use the web optimizer today, is something going on?
IE - no sign in button
Chrome - new build button does nothing
Firefox - new build button does nothing
side note, first time ever seeing a 416 error, so there is that.
Make sure you're logged in with your ArcGIS Developers account. An AGO or global esri acct won't cut it and there's no indication on the page to let you know that unfortunately.
