I want to manipulate on points, which is in particular layer

I am not ArcPy developer, so may I have a dumb question, but I will try. In ArcMap, I want to add a specific value in the column for point, which is in the selected layer. For example, someone will select the layer and I want to run for loop and change values in db for the points which is in that layer. Is it possible? What can I do?

I might be missing the details of what you want to do, but it sounds like after selecting a layer you could either call the Calculate Field tool or run through rows with an Update Cursor.
