Is there a way to unclock the "supports calculate" attribute in a Featurelayer?

??? What do you mean. As the documentation states this is only an option for hosted services on AGOL currently.

What about the rest service? how to active that?

Evelyn,

There is no way that I am aware of. If the layer has editing enabled then this is enabled for hosted layers.
