Could you please provide more details on your issue? What is the error message that pops up? Try opening your browser's developer tools and going to the network tab to see what the request to upload the manifest.json returns.

A registered folder is not a webserver.

The steps for "Host a custom widget on a web server" will guide your through the setup to enable Portal to access the server. You will need to set up a machine that is configured within your ArcGIS Enterprise.

There's no link in your post.

The manifest.json needs to be on a webserver that is a part of your enterprise (i.e. accessible to your portal server via HTTPS).
