1. I have finished  import S-57 files into a geodatabase and added the Maritime.lyrx 

or

2. Create a tile package using Maritime Chart Service（ run the mcstpk.exe ）

You can use either method. If you use the maritime chart service and the mcstpk.exe, you do not need to import the S-57 to geodatabase. It might help to know a little bit about your intended use of the tile package to suggest one method over the other.
