How to change the color of a polygon in ArcPy?

I am trying to write a script that changes the color (fill and outline color) of a single polygon in a layer using ArcPy. Is this possible without importing properties from a reference layer file using arcpy.mapping.updatelayer()?

The only way I know of is to use Layer—Help | ArcGIS for Desktop files.
