We're experiencing a strange phenomenon with TPKX files in the layout. When zooming in the layout, the pixels are blended into larger pixels. It seems like an inverse rendering is occurring. Even if you export the layout to a PDF, for example, the result is just as poor. We didn't have this problem in an earlier version of AGP (we're trying to research the version).
When you are zooming in on the layout, it's zooming in to the layout page not the map. The layer tiles don't get updated unless you change the scale of the map in the layout. Could that be what's going on here?
A nice idea, but it doesn't affect the display and output quality, which is significantly worse. Changing the scale wouldn't solve the problem either. We didn't have this problem in earlier versions of AGP. We're still looking to see if we have an older version on a computer somewhere. Please try it yourself to see if you can reproduce the problem.

I don't think I'm understanding the situation fully. It sounds like something displayed differently in an earlier version, and that export quality is now different. With the screenshot I can't speak to the export quality, and I'm not sure what you mean about zooming into the layout.
I can try with my own tile package layer, but there are a lot of parameters you can adjust in a tpkx, any of which could play a role in this. With different files and parameters I may not see the same issue. If you can I recommend reaching out to tech support as they have the resources to debug this and get it to the right development team to address. They may also have a work around or best practice recommendations.
