Good Afternoon:

Question 1: Can Portal be configured to search Geo-referenced data from MS Word, PowerPoint, etc. ?
Question 2: If Yes, is there a standard format that the MS Word, PowerPoint, etc, has to be in?

I am a bit confused by your first question.  What do you mean by "search Geo-referenced data"?  Are you asking if Portal can include MS Office documents in its search results just as with geographic content or are you asking if Portal can literally search inside an MS Office document stored in Portal or are you asking something else?  Generally speaking, users can upload and share MS Office documents from within Portal.  This section of the Esri help discusses that functionality a bit. 

I'm also confused by your second question.  What do you mean by "standard format"? 

On the other hand, you might be asking if users can search Portal's geographic content from within MS Office applications.  If that's the case, then the answer is 'yes, somewhat'.  Esri Maps for Office loosely integrates MS Excel and MS PowerPoint with Portal for ArcGIS in that you can create maps from shared content on Portal or uploaded content.  This functionality is not available in MS Word.

With respect to Question 2: When hosting a Word doc on Portal, is there a way to tag the document so it can be searched by keyword or geographic extent?

Thanks very much for clarifying.  Regarding question 1, I believe the answer is 'no'.  Searching Portal for content will only return a result set based on specific details about the document such as owner, title, type, description, etc.  In other words, Portal cannot scan inside of a document to find a string or parse it as you described.  Regarding question 2, yes absolutely... in fact, tags are one of the best things you can do with your content because it allows you to associate keywords which helps to facilitate Portal's search functionality.  Creating tags can be accomplished through Item Details for a particular document, map, layer, etc. that you have uploaded to Portal.  Clicking the Edit button within the Item Details section allows you edit the title, summary, thumbnail image, description, access and use constraints, tags, credits, and extent.  You can add tags to Items and to Groups.
