Is it possible to bring a layer I have drawn in Illustrator into my ArcGIS online project?

I would recommend you to draw the polygons in ArcGIS in the first place. If you take them out of Illustrator with the objective of calculating the area you need to make sure that you export georeferenced data in order to get correct measurements. I am not a user of Illustrator so I do not know what the options are.
