You'll need to use some format tags that are available. Read about Formatting tags available in ArcMap.

to get E=mc²

I think you would need to enter "E = mc" & "<SUP>" & "2" & "</SUP>

Using <SUP> </SUP> around the text that is meant to be the superscript.
