Calculate Slope Between Points

I've been trying to find some threads on calculating slope between points, but I've only had luck finding slope using rasters.

I have a drainage line that was created using CAD and I brought it in to ArcMap and overlayed it on my DEM. I created points at 100m intervals along the drainage line and then converted those points to 3D to get the xyz values. I was then asked to create perpendicular lines from the drainage line at given lengths - say 100m on each side of the drainage line. At the ends of the perpendicular lines, I have the xyz values.

What I need to find next, is the slope from the end of the perpendicular lines to the next point (100m away) along the drainage line. I will need to repeat this process along the entire length of the drainage line. I've attached a screen shot to show you what I'm after.

Is there a tool that can calculate this, or a process that I'm overlooking to determine the slope? In all honesty, I'm not really sure where to begin.
