I have a DEM in normal raster format and a polyline feature class of a river. I am attempting to extract the elevation values from the raster in the cells that contain the river. Is this possible and how can I achieve this?

You should either convert the raster into a feature polygon/point or convert the river feature to a raster. In feature class you can you spatial join / intersect. In raster format you can use raster 0*1 (river=1 and rest =0) multiplication to get your desired values.
