I am trying to publish a service to my Hosted Layers within AGO from Desktop and its not giving me access to My Content when running publish service wizard. Do I not have Publisher rights with a basic desktop license? thanks
ArcGIS Desktop Basic should be able to publish a hosted feature service to ArcGIS Online.

Do you have the appropriate permissions in your ArcGIS Online account?

Login to your AGOL organization, then go to your Profile (you can access from the dropdown list in the upper right).
On the 'My Profile' page, check your role: it should say 'Publisher'.
You could also ask your AGOL admin as Katie Cullen suggested in her post.
Contact the administrator in your organization to ensure that your account has publishing privileges
