sampling a grid using sample points as centers of buffers

My goal is quantify the landscape in circular buffers around 100+ points.  These are points where biological data were collected and I want to see if there is a relationship between the biological data and the landscape data. The grid is the NLCD land cover map. In the past I used raster calculator to (1) create binary maps of each land cover type (e.g., urban, crop, etc) and (2) sample the land cover map with circular buffers.  My previous issue was raster calculator couldn't recognize my shapefile and my latest issue is that raster calculator does not recognize the raster (transformed from the shapefile).  Code below.

I was wondering if there is a tool that would accomplish my goal using the original map or the binary maps extracted from it.
