I have an issue when publishing from a registered folder to an enterprise portal, but it only allows me to publish map images only, and when I select the feature layer an error message says that  "layer's data source is not supported (web feature layer)"

That’s by design.  You have to use an enterprise geodatabase.  Files can only be edited by one process at a time, a windows limitation.  1 SOC is one process.  More than 1 soc would equate to many users.

Just curious, why do you want to enable 'feature layer' while publishing data, I assumed, off a file gdb? Is your goal to allow editing from web applications? Or just drawing on the client app using some web clients' advanced symbology option? or something else? thanks
