This is one example of where you need and Enterprise Geodatabase (Oracle, SQL Server, Postgres) so that you can update the mosaic database while it is being used. The alternative is to add the new rasters to a copy of the original and when ready, shutdown the server and publish the updated mosaic dataset, which can be done very quickly.
