Is there any Idea , what i can do:

C:\Program Files\ArcGIS\DataStore\tools>configuredatastore http.. usr pwd d:\datastore\ --stores tileCache
Data Store wird konfiguriert....
Error encountered: Vom Computer 'https://localhost:2443/arcgis/datastoreadmin' .....
Extended error message: Upgrade for Data Store vom Typ "tileCache" not done..
Extended error message: CouchDB failed to start"

Yes.

See Esri BUG-000116596 ("Unable to configure tile cache data store if port 5986 is utilized.").

WinRM und CouchDB are using the same port.
