I want to use extract data function in my web application, Is it possible to use SpatialAnalysisTools function without arcgis portal?

If you want to use the standard Spatial Analysis tools, you'll need an ArcGIS Enterprise deployment, (Portal, hosting server, registered ArcGIS Data Store). If you're interested in extracting data, you can look into the Clip and Ship geoprocessing service. This requires you to author the service with specific data to clip, though.

I do not have Portal installed at least to my knowledge.

This isn't so much a question about IIS. Yes, you'll need it for the web adaptors, but once you get a base deployment set up, you should be able to use the Spatial Analysis tools.
