It depends on how you configure Portal for ArcGIS. If you decide to enable some of the utility services, such as geocoding, directions and routing, and geoenrichment - then yes, it can use credits.

Configure utility services—Portal for ArcGIS (10.6) | ArcGIS Enterprise 

Alternatively, you could use your own utility service (e.g., geocoding service) and not use credits.

HI Derek,

I understand that using extra ordinary tools and such would use your AGO credits, but what about hosting services and data in your Portal does that use credits, because AGO charges you credits monthly based on how many gb of data are stored in your content.?

For storing hosted services and data in a Portal for ArcGIS deployment - configured as a hosting server (aka. ArcGIS Enterprise base deployment), no credits are consumed.
