Centerline is only a geometry line in the UPDM database, it should not have any measure value. Is it right?

Yes, centerline feature class does not store measure values. Centerline feature class is a polyline feature class that stores the route geometry. This centerline geometry, along with route definitions, are stored in a network feature class which is polyline M enabled. Calibration point feature class stores the route measure values. For in-depth understanding on data model please refer - http://desktop.arcgis.com/en/arcmap/latest/extensions/pipeline-referencing/alrs-data-model.htm#ESRI_...

Thanks, Anjali. if the centerline is M enabled, is there any potential issue for the database?

As documented, the centerline feature class should be Z enabled but not be M enabled. Since it is recommended that the Centerline Feature class not to be M enabled, we have not tested tools to see potential places that it might have issues. Also, user will experience that the tool will not take M enabled centerline feature class as an input.
